From mass spectrometry to MALDI imaging

Imaging mass spectrometry

Imaging mass spectrometry used in the JCSM allows the examination of tissue sections and thus the exact localization of thousands of proteins in an experiment. The advantage over previous proteomic studies is that the proteins need neither be labeled nor known beforehand. Using this method, the heterogeneity of tumors can be detected due to the different distribution of proteins but also of lipids. It is to be expected that with the help of this novel method also new tumor markers can be found.

Geräte

ESI/APCI-TOF Massenspektrometer

The micrOTOF QII is a Bruker Daltonics time-of-flight mass spectrometer. The instrument can be operated with either an electrospray ionization (ESI) source to analyze plarious substances or atmospheric pressure chemical ionization (APCI) for the analysis of nonpolar substances. By coupling to an HPLC system (Agilent 1200 series with diode array detector), sample components can be additionally separated before mass spectrometric detection.

MALDI-TOF/TOF Massenspektrometer

The Ultraflex III uses matrix-assisted laser desorption / ionization (MALDI) in a tandem mass spectrometer. Different methods of measurement allow both the analysis of large molecules in linear mode and the detection of smaller molecules with greater mass resolution and accuracy through the use of a TOF reflector in reflector mode. The device also has a LIFT unit for MS / MS analysis.

Gaschromatographie - Massenspektrometrie

In addition, our labs are equipped with two Shimadzu GC-MS systems (GC-2010 / GCMS-QP2010), which contain universal low-polarity columns (diphenyldimethylpolysiloxane), but allow identification of analytes using a quadrupole mass spectrometer.
